Chloramphenicol(CAP)Rapid Detection Card——User manual
Chloramphenicol detection card is used for rapid detection of Chloramphenicol residues in aquatic products(fish,shrimp,crabs,etc.),livestock and poultry meat,and sediment in water bodies
The entire detection process only takes about 30 minutes.This test strip is a qualitative detection,and the sensitivity of the detection card is 0.1~0.3ppb.
1、[Detection Principle]
The chlorine detection card applies the principle of competitive inhibition immunochromatography.During the flow process,the chlorine in the sample binds to a specific monoclonal antibody labeled with colloidal gold,inhibiting the binding of the antibody to the chlorine protein conjugate on the NC membrane detection line(T).If the chlorine content in the sample exceeds the detection limit,the color of the detection line(T)is lighter than that of the C line,and the result is positive;On the contrary,if the color of the detection line(T)is darker or the same as the quality control line(C),the result is negative.
2、Product composition
Self provided equipment and equipment are required:
Stirrer;Oscillator;Electronic balance;Centrifuge;Air drying instrument;Pipette;
15mL centrifuge tube(for centrifugation);5mL centrifuge tube(for blow drying)
3、Sample processing
1.Peel and fat fish,shrimp,crabs,livestock and poultry meat,etc.,homogenize the sample with a homogenizer,take 2g of fat free tissue sample,and add it to a 15ml centrifuge tube;
(Take 2g of sediment and sediment samples and add them to a 15ml centrifuge tube);
2.Accurately add 4mL of extractant to the centrifuge tube,tightly seal the bottle cap,vigorously shake for 3 minutes,and centrifuge at 4000rpm for 5 minutes;
3.Use a pipette to pipette a certain amount of supernatant(as shown in the table below)into a 5ml centrifuge tube,and blow dry with a blow dryer at 65℃;
The relationship between the amount of supernatant removed and the detection limit is as follows:
4.Add 0.3ml of the reconstituted solution to the dried centrifuge tube,and dissolve the solid residue at the bottom and inner wall of the cup.This solution is the test solution.
4、[Usage Steps]
1.Before conducting the test,please read the user manual in its entirety.Before use,restore the test card and sample solution to room temperature(20-30℃);
2.Please use the testing card as soon as possible after removing it from the packaging bag;
3.Place the detection card flat and take 80µl of the sample to be tested into the sampling well;
4.Timing starts when the liquid flows,and after 5-8 minutes,the result is determined based on the schematic diagram.Other time readings are invalid(when reading the result,place the detection card horizontally in front of the observer).
5、【Result Interpretation】
1.Negative(-):The color of the detection line(T)is darker or the same as that of the quality control line(C),indicating that the medium concentration of the sample is below the detection limit or does not contain chlorine.
2.Positive(+):The detection line(T)shows a lighter color than the C line,or the detection line(T)does not show a color,indicating that the residual concentration of mycomycin in the sample is greater than the detection limit.
3.Invalid:The quality control line(C)does not show color;It may be due to improper operation or detection card failure.Use a new detection card to retest.
6、【Precautions】
1.This pre-treatment can be used in conjunction with our company's rapid quality and safety testing box for aquatic products,and an experiment can be completed without the need to purchase other instruments.If you need relevant information,please contact our company.
2.Disposable detection card;Do not use expired detection cards,and dispose of waste properly;Do not touch the white film surface in the center of the detection card;Do not reuse the pipette suction head to avoid cross contamination;Do not consume the reagents provided.
3.When conducting sampling and testing operations,it is recommended to use live fish,live shrimp,or fish and shrimp that have been frozen for no more than two weeks as much as possible.Shrimp needs to undergo experiments after turning and shell removal,while fish need to remove scales and take parts of the fish's back that have less oil for experimentation.To ensure the accuracy of the experiment,it is advisable not to take parts of the fish's belly and tail that have more oil for experimentation,as more oil will adhere to the membrane surface of the observation area and affect the lateral chromatography system,interfering with the accuracy of the experimental results.
4.Do not mix reagents and test cards from different batches.
7、[Storage and expiration date]
Store in a cool and dry place at 4-30℃,away from light;The validity period is 12 months(production date and batch number can be found in the packaging box).
